Development of a Lateral Flow Assay for Bevacizumab Using an Anti-Idiotype DNA Aptamer as a Capture Molecule

We developed a point-of-care testing method based on lateral flow assay for bevacizumab, using an anti-idiotypic DNA aptamer as the capture molecule. Bevacizumab loaded onto sample pad strip bound to red-colored aptamer-modified gold nanoparticles (AuNPs) in conjugation and then migrated into developing zone. The AuNPs bevacizumab were trapped by protein A test spot. In contrast, excess amounts of unreacted control spot, confirming that they correctly. strips enabled analysis within 20 min after adding sample, was quantified photographing colored areas with smartphone analyzing images. kit capable quantifying range 0–200 μg/mL Avastin diluent bevacizumab-spiked human serum samples. This enables on-site concentrations simple operation without special analytical apparatus.
